In a humongous interview with IGN, Rockstar kingpin Dan Houser has described Red Dead Redemption's multiplayer as "charging around on horses and stagecoaches and trains, shooting each other". So basically, it's GTA4, but with more dust and tumbleweeds, presumably racing to deliver packages of Dr Hobbton's Famouse Frontier Coughe Linctus instead of cocaine.
He's also dropped some details on the story gameplay. "You'll have a load of missions open at any one time, so you'll have the choice of 'do I go and do a mission for him or do I go and do a mission for them'…There's a couple of points when you have choices that you can make in that story but not that much…Our main goal with a game like this or with any game is giving the player freedom over what they do next. There's always a ton of other activities, both mission-based and non mission-based to go and do." Soooo ... it's GTA4, but with more dust and tumbleweeds. Again.
